Adipose Tissue Stromal Vascular Fraction Cells Differentiate Depending on Various Methods

Abstract
Objective: Adipogenic medium of fat cells can be induced using culture medium supplemented with insulin, 3-isobutyl-methylxanthine (IBMX), glucocorticoid and indomethacin. The purpose of this study was to determine medium conditions for promoting differentiation of stromal vascular fraction (SVF) cells towards adipocyte.

Methods: SVFs were isolated subcutaneous adipose tissues and retroperitoneal adipose tissue of Long-Evans Tokushima Otsuka (LETO) rat and Sprague-Dawley rat. Post-confluent SVFs were cultured MDI medium (520 ¥ìM 3-isobutyl-methylxanthine (IBMX), 1 ¥ìM dexamethasone, 1 ¥ìg/mL insulin) for 2 days 3 times or MDII medium (MDI medium, 200 ¥ìM indomethacin) for 3 days 3 times or MDIT medium (MDI medium, 20 ¥ìg/mL troglitazone) for 2 days.

Results: SVF cells adipogenic differentiation were higher to add the MDI medium 3 times, MDII medium or MDIT medium than add the MDI medium.

Conclusion:Adipogenic differentiation was enhanced adding the adipogenic medium multiple times and using the different PPAR¥ã ligands (indomethacin, troglitazone).
